Abstract :

From the library of Klebsiella variicola T29A mutants, it was observed that the mutant strain 9:20, T29A2 - miniTn5Km of Klebsiella variicola T29A, which was disrupted in a gene that was denominated fxn9:20 (for its subsequent identification), showed sensitivity to desiccation for stress. Bacterial desiccation is the ability of an organism to lose intracellular water until it reaches equilibrium with the water present in the air. The objective of this work is to determine the sensitivity of the 9:20 mutant strain of K. variicola T29A in environmental and controlled desiccation (lyophilization) conditions. K. variicola T29A is a symbiotic bacterium isolated from the rhizosphere of sugarcane plants which has the ability to promote plant growth in sugarcane plants by mechanisms such as nitrogen fixation. To determine the sensitivity to desiccation, the bacterial survival rate (BSR) was calculated for the mutant and wild type strains. It was observed that with the environmental desiccation, the BSR decreases as the stress time increases. After 30 days of desiccation stress, the wild type bacteria had a BSR=84.88 and the 9:20 mutant strain did not show growth with a BSR=0; this result proves the sensitivity of the mutant bacterium. The main objective of this project is to characterize the mutated gene and to determine its function in the desiccation tolerance of Klebsiella variicola.
